摘要
Objective: To establish a chronic periodontitis model of SD rats and detect the expression of Notch in peripheral blood mononuclear cells (PBMCs) of chronic periodontitis model was detected by real-time quantitative polymerase chain reaction (RT-PCR), intracellular staining and flow cytometry. It is hoped that we can find out the potential significance of Notch related molecules in the development of chronic periodontitis and provide a theoretical basis for seeking new methods of treating chronic periodontitis. Methods: 27 male SD rats aged 6-8 weeks were randomly divided into control group and model group. By means of Porphyromonas gingivalis, a chronic periodontitis rat model was established. The gingival index, tooth mobility and periodontal pocket depth were recorded. The expression levels of Notch signaling related receptors (Notch 1 to Notch 4) in PBMC of model mice were detected by fluorescence quantitative polymerase chain reaction, flow cytometry and intracellular staining at different levels and compared with the control group. Results: The model of chronic periodontitis in SD rats was successfully established. The expression of Mrna of Notch 1, Notch 3, Notch 4 and percentage of cell expression in the model group were significantly lower than that in the control group (P<0.01), but Notch 2 Mrna and percentage of cell expression were significantly higher (P<0.05). Conclusion: This indicated that Notch and its ligands may play important roles in imbalance of immune balance for chronic periodontitis and provide ideas for in-depth study of the pathogenesis of chronic periodontitis.
