Effects of inflammatory response and oxidative stress on the GLUT-1 signal transduction in the placenta of patients with gestational diabetes mellitus

摘要 Objective:To investigate the effects of inflammatory response and oxidative stress on the GLUT-1 signal transduction in the placenta of patients with gestational diabetes mellitus. Methods: A total of 102 patients with GDM who were treated and gave birth in our hospital between December 2014 and August 2017 were selected as GDM group and 100 normal puerperae who gave birth in our hospital during the same time period were selected as normal control group. The differences in the expression of GLUT-1 gene in placental tissue as well as the contents of inflammatory factors and oxidative stress indexes in the placental grinding liquid were compared between the two groups. Pearson test was used to evaluate the effects of inflammatory response and oxidative stress on the GLUT-1 signal transduction in the placenta of patients with GDM.Results: Inflammatory factors IL-2, TNF-α and LEP contents in placental grinding fluid of GDM group were higher than those of normal control group whereas APN content was lower than that of normal control group;oxidative stress index LPO content was higher than that of normal control group whereas GST, CAT and SOD contents were lower than those of normal control group;GLUT-1 mRNA expression in placental tissue was lower than that of normal control group. Pearson test showed that the inflammatory response and oxidative stress levels in the placenta of puerperae with GDM were directly correlated with the GLUT-1 expression.Conclusion: There are significant inflammatory response and oxidative stress response in the placental tissue of puerperae with GDM, which can directly decrease the GLUT-1 gene expression and inhibit the GLUT-1 signal transduction.
