基于生物分子网络调控研究麻黄附子细辛汤治疗过敏性鼻炎的作用机制 被引量:16

Mechanism of Mahuang Fuzi Xixin Decoction in treating allergic rhinitis based on biomolecular network regulation

摘要 目的 基于生物分子网络研究麻黄附子细辛汤治疗过敏性鼻炎的作用机制.方法 利用中药系统药理学数据库和分析平台(TCMSP),筛选口服生物利用度≥30%和药物相似性≥0.18的成分,获得麻黄附子细辛汤的有效成分组.利用UniProt数据库对麻黄附子细辛汤的有效靶点进行名词统一,并利用比较毒物遗传学数据库(CTD)进行靶点相关疾病注释,得到抗过敏性鼻炎相关靶点.将上述靶点输入到String数据库进行蛋白质-蛋白质相互作用网络预测,最后利用DAVID数据库进行基因本体(GO)与KEGG富集分析,揭示麻黄附子细辛汤治疗过敏性鼻炎的潜在信号通路.结果 筛选出麻黄附子细辛汤的35个活性成分和112个抗过敏性鼻炎靶点,其抗过敏性鼻炎作用可能与人肿瘤蛋白P53(TP53),转录因子AP-1(JUN),白细胞介素6(IL-6),血管内皮生长因子(VEGFA)和肿瘤坏死因子(TNF)等有关.此外,麻黄附子细辛汤抗过敏性鼻炎的靶点显著富集于钙信号通路,核苷酸结合寡聚化结构域(NOD)样受体信号通路,T细胞受体信号通路,丝裂原活化蛋白激酶(MAPK)信号通路,Toll样受体信号通路,细胞因子-细胞因子受体相互作用等信号通路中.结论 麻黄附子细辛汤能够多靶点、多途径、多通路地调控生物分子网络,其抗过敏性鼻炎的潜在生物学机制可能与钙信号、NOD样受体、T细胞受体、MAPK、Toll样受体等信号通路有关. Objective To study the mechanism of Mahuang Fuzi Xixin Decoction(MFXD)in treating allergic rhinitis.Methods The effective components of Mahuang Fuzi Xixin Decoction were obtained by using TCMSP database(oval bioavailability≥30%and medical similarity≥0.18).UniProt database was used to unify the terms of effective targets of MFXD,and comparative toxicology genetics database(CTD)was used to annotate target-related diseases,and as a result,MFXD anti-allergic rhinitis related targets were obtained.The above targets were input into the String database for protein-protein interaction network prediction.Finally,gene ontology(GO)and Kyoto gene and genome encyclopedia(KEGG)enrichment analysis were carried out using the DAVID database to reveal the potential signal pathway of MFXD in the treatment of allergic rhinitis.Results 35 active ingredients and 112 anti-allergic rhinitis targets of MFXD were selected.Its anti-allergic rhinitis may be related to human tumor protein P53(TP53),transcription factor AP-1(JUN),interleukin><385F>6(IL-6),vascular endothelial growth factor(VEGFA)and tumor necrosis factor(TNF).In addition,anti-allergic rhinitis targets of MFXD significantly enriched in calcium signaling pathways,nucleotide combined with structure of oligomerization domain(NOD)receptor signaling pathways,T cell receptors signaling pathways,mitogen activated protein kinase(MAPK)signaling pathway,toll-like receptors signaling pathways,such as cytokines,cell factor receptor interaction signaling pathways.Conclusion MFXD can regulate biomolecular networks with multiple targets and multiple pathways,and its potential anti-allergic rhinitis,multiple pathways biological mechanism may be related to calcium signaling,nod-like receptor,t-cell receptor,MAPK,toll-like receptor and other signaling pathways.
