摘要
目的:从血清蛋白质组初步探讨艾灸免疫调节中蛋白质分子的变化规律。方法:采用超速离心除盐方法预处理血清,进行一维和二维聚丙烯酰胺凝胶电泳(2-DE)分离,凝胶银染显示各组蛋白质差异表达。结果:2-DE图谱中可辨识的蛋白质斑点达300~400个,除有看家蛋白、特异蛋白外,不同时段存在差异表达蛋白,从十几个至几十个不等。艾灸经穴组的蛋白质组分变化较为明显,以酸性蛋白点缺失和蛋白峰度下降最为主,而非经穴组血清蛋白变化不明显。结论:在特定穴位上艾灸刺激有助于清除血清中的免疫抑制物,使艾灸血清的蛋白质组分发生以酸性蛋白缺失为主的改变。
Objective:To study the changing rules of protein molecules for immunological regulation by moxibustion from the viewpoint of serum proteome. Methods: The serum was prepared by clearnning up by removing salt,and then separated by one and two demensional electrophoresis. Differential proteins in various groups were detected on silver stainning gels. Results:In each group,the numbers of cognizable protein dots ranged from 300 to 400. Apart from house-keeping proteins,each group has its own special proteins, and they varied from time to time. The numbers of differential proteins were up to a decade or several decades. In the course of moxibustion, distinct transformation occurs in serum proteins particularly in disappearance of protein and declining of protein kurtosis. It did not happen on non-acupoints group. Conclusion:Moxibustion at specific acupoints helps to remove immunosuppression substances,especially the acid proteins in proteome.
