摘要
目的探讨铸铜石英砂体外致脂质过氧化作用及讨枸杞多糖对其的拮抗作用,为矽肺的防治提供新的途径。方法于小鼠肺组织匀浆中加入不同剂量的铸铜石英砂单独培养及同时加入铸铜石英砂和不同剂量的枸杞多糖共同培养后,分别检测各组的羟基自由基(-OH)及谷胱甘肽过氧化物酶(GSH-Px)。结果不同剂量的铸铜石英砂在小鼠肺组织中能产生-OH且可抑制GSH-Px酶的活性,产生-OH的能力和抑制GSH-Px酶活性的作用均有随铸铜石英砂染毒剂量的改变而改变的趋势;各剂量的枸杞多糖能抑制-OH的产生,其中抑制水平以0.25 mg/g剂量组最大;而提高GSH-Px酶活性的作用则见于不同剂量的枸杞多糖,且以0.50 mg/g剂量组作用最强。结论铸铜石英砂在体外具有致脂质过氧化的作用,枸杞多糖可拮抗该作用,提示可将枸杞子作为防治矽肺的药用或食用品。
Objective To study the lipid peroxidation of cast copper quartz sand in vitro and explore the antagonism of medlar amylose on the lipid peroxidation of cast copper quartz sand.Methods Different doses of cast copper quartz sand were added into mice lung homogenate and cultured.,and different doses of cast copper quartz sand and different doses of medlar amylose were simultaneously added into mice lung homogenate and cultured.After incubation,the contents of-OH and GSH-Px were determined respectively.Results Different doses of cast copper quartz sand could bring out-OH and inhibit the activity of GSH-Px.The ability to bring out-OH and the inhibition of GSH-Px varied as the doses of cast copper quartz sand varied.Each dose of medlar amylose could inhibit the generation of-OH with the strongest actions in the 0.25 mg/g group.The increase of GSHPx activity could be observed in each dose of medlar amylose with the strongest actions in the 0.5 mg/g group.Conclusion Cast copper quartz sand could bring out lipid peroxidation,and medlar amylose could antagonize such actions of cast copper quartz sand in vitro,which suggests that medlar amylose could serve as the medicine or food against silicosis.
