摘要
目的:探讨钩藤碱(Rhy)对人体肠系膜动脉平滑肌细胞大电导钙激活钾通道(BKCa)的作用。方法:切取人体肠系膜动脉并分离细胞,以膜片钳测BKCa活性。结果:(1)高血压病组(EH)与非高血压组(NH)比较,前者BKCaPo较高,Tc较低;加入Ca2+后,与加Ca2+前相比NH组Po的增加幅度高于EH组。(2)加入Rhy后,在细胞贴附式BKCa无反应;在内面向外式EH组BKCaPo增加的幅度高于NH组,同一浓度下,前者Po较高,Tc较低。结论:(1)EH组BKCa活性高于NH组。(2)Rhy可直接激活BKCa,且在EH组更为显著。
Objective:To study the regulation of Large-conductance calcium-activated potassium channels(BKCa)of mesenteric artery smooth muscle cell by rhynchophylline(Rhy).Methods:The mesenteric artery branches from essential hypertention(EH)and non-hypertention(NH)were digested by enzyme into single cells.Patch clamp technique was used to record the Open Probability(Po),Amplitude(Am),Mean Open Time(To) and Mean Close Time(Tc).Results:(1)Compared to NH,the Po of EH was higher and the Tc was lower.Adding Ca2+,the Po increasing of NH was more obviously than EH.(2)Adding Rhy,the Po increasing of EH was more markedly than NH.At the same concentration,the Po of EH was higher and the Tc was lower than NH.Conclusion:(1)The BKCa activity of EH was greater than NH.(2)Rhy could open BKCa directly,and the effect on EH was more distinct than that on NH.
