高效液相色谱-质谱联用法分析冬虫夏草及其功能食品中的活性成分 被引量:16

Determination of Active Components in Cordyceps Sinensis and Its Functional Food by HPLC-MS

摘要 目的:分析冬虫夏草及其功能食品中的主要活性成分。方法:选择不同溶剂提取冬虫夏草样品,比较它们的提取效果;采用高效液相色谱-电喷雾质谱结合选择离子监测(SIR)法分析冬虫夏草中虫草酸、虫草素、尿嘧啶等活性成分。结果:确立了样品提取的最佳溶剂;经色谱、质谱条件的优化,建立了定量分析冬虫夏草及其功能食品中活性成分的方法。虫草酸、尿嘧啶、次黄嘌呤、尿苷、腺嘌呤、腺苷、虫草素的定性离子分别为183、111、137、265、136、268、252,检出限(LOD)分别为74.2、72.0、85.0、82.0、4.2、50.0、65.0ng/mL(进样量5μL),回收率90%~110%。结论:该方法灵敏度高、选择性好、操作简便、定量准确,适用于冬虫夏草及其功能食品中的活性物质分析及产品质量控制。 Objective:To determinate main active components(cordycep acid,uracil,hypothanthine,uradine,adenine,adenosine and cordycepin)in Cordyceps sinensis and its functional food.Method:Different solvents was selected to extract active components in cordyceps sinensis sample and compared the effect of extracting solvents on extracting ratio.The active components of Cordyceps sinensis were determined by HPLC combined with electrospray mass spectrum(HPLC-ESI-MS).Result:Water was chosen as extracting solvent.HPLC-MS method with SIR mode was established to determinate main active components.Qualification ions of cordycep acid,uracil,hypothanthine,uradine,adenine,adenosine and cordycepin were 183,111,137,243,136,268,252,respectively,limits of detection were 74.4,72.0,85.0,82.0,4.20,50.0,6.05 ng/mL,respectively(inject volume 5 μL).Recoveries were between 90%~110%.Conclusion:This method showed high sensitivity,accuracy and easy to perform.It is suitable to analysis active component in Cordyceps sinensis and its functional food,as well as quality control of product.
