摘要
随着基因工程及分子生物学的快速进展,对检测基因所必需的探针及其应用研究已提到日程上来。为此制备国产的长臂生物质及光敏生物素已作为国家重大课题进行研究。鉴于同位素探针的高度敏感性及特异性,其在国内、外的有关工作中至今仍占有相当的比重。我们在制备^(32)P HBV DNA探针诊断药盒的过程中,对国产的和英国的α-^(32)P-dATP进行对比试验。
HBV DNA probes, labelled by α-^(32)P-dATP from England Amersham International PIC and from China Institute of Atomic Energy, were compared in specific activity, sensitivity, incorporated percentage and stability in different temperatures for storage. We obtained almost the same satisfactory results for the two probes. HBV DNA were detected in 551 and 518 samples sera, collected in hepatitis laboratory(u=1.0056 ,P>0.05).
