壳聚糖酶高产菌株的筛选及酶解产物的定性 被引量:8

摘要 采用平板透明圈法 ,从 60份采自全国各地的土样中筛选到了 45株产壳聚糖酶菌株。以Imoto法测定酶解所产生的还原糖量高低来判定产酶能力 ;对产酶能力较高的菌株PZ2 1 -3 ,CJ2 2 -3 ,用HPLC、MS和TLC法检测酶解产物 ,确定其产内切酶。对CJ2 2 -3进行初步鉴定为曲霉。 Forty-five strains produce chitosanase which could form a clear zone around the colony with chitosanase detection agar plate, were isolated from sixty soil sample. Imoto method was used to determine enzyme activity.HPLC and TLC were used to analyze hydrolysis products. It was indicated that strain CJ21-3 and strain CJ22-3 had endo-chitosanase and strain 22-3 was identified as a member of Aspergillus.
