摘要
我们的前期研究发现运动性肥大心脏心肌血管紧张素Ⅱ(angiotensinⅡ,AngⅡ)受体数目减少,但对这种变化的调控是否可发生在基因转录水平上目前尚不清楚。本实验采用反转录-聚合酶链式反应(ReverseTranscription-PolymeraseChainReaction,RT-NR)方法对运动性肥大心脏心肌AngⅡAT1受体工型mRNA表达进行研究。结果表明大鼠游泳运动8周后,心系数比对照组大鼠提高24%(p<0.05)。琼脂糖电泳结果表明心肌AbgⅡAT1受体亚型RT-PCR产物条带密度比对照组降低30%,面积降低61%(p<0.05)。说明运动性肥大心脏心肌AngⅡAT1受体基因表达降低。提示运动性肥大心脏心肌AngⅡ受体数目减少的调控机制可发生在基因转录水平上。
Our previous studies showed that the number of angiotensinⅡ (Aug Ⅱ ) receptors was decreased in exercise - induced hypertrophied rat heart. However it is still not clear that number decrease of Ang Ⅱ receptor is due to downregulation in its gene expression.
