摘要
用人IgG-Sepharose 4B亲和柱,3mol/L氯化镁洗脱,一步纯化免抗人IgG血清,获得了SDS-PAGE电泳纯,免疫双扩散法效价达1/256,收率62.6%;用8mol/L脲洗脱,效价为1/128,收率34.1%.与硫酸铵多步沉淀法比较,纯度和效价大大提高,为获得高度特异、高亲和力兔抗人IgG提供了一条简便途径.
Using humanlgG-sepharose 4B affinitychromatography column with 3 mol/L MgCl2 eluant, the rabbit anti-human IgG antisera were isolated in one step. The results revealed that the rabbit anti-human IgG was pure in SDS-PAGE, its titer was 1:256 in IDD, the yield was 62. 6% . Replacing 3 mol/L MgCl2 with 8 mol/L urea, the titer in IDD was 1:128, the yield was 34.1%.
