褪黑素对苯妥英致畸效应的拮抗作用

摘要 背景与目的 :探讨褪黑素(MT)对苯妥英(DPH)诱发小鼠的致畸效应的影响。材料与方法 :于母鼠妊娠GD6 -15 每日灌胃染毒1次不同剂量的MT、DPH ,或两者合并给药 ;GD18 解剖检查 ,观察各项指标。结果 :各实验组在平均活胎数、死胎率、平均着床数、窝平均黄体数及吸收胎数等指标上未见明显差异。高、低剂量DPH组畸胎率分别为18.20 %和45.03 % ;明显高于溶剂对照组。MT与低剂量DPH共处理可明显降低DPH的致畸效应(5 %~12.24 %)。此外 ,MT对低剂量DPH诱发的胎仔体重和身长降低也具有拮抗作用。结论 BACKGROUND&AIM: To test the hypothesis that antioxidant melatonin(MT)can reduce the incidence of phenytoin(DPH)induced teratogenesis. MATERIAL AND METHODS: Female Kunmin mice were administered by gavage with DPH,MT or DPH and MT simultaneously at GD 6-15 after coitus and subjected to caesarean section for various examinations.
