半边莲通过钙信号诱导肝癌细胞凋亡的实验研究

Experimental Study of Hepatoma Cells Apoptosis Induced by Lobelia Chinensis Lour.through Calcium Pathway

观察半边莲通过影响胞内游离钙离子浓度进而诱导人肝癌细胞HepG2发生凋亡。通过荧光分光光度法检测胞内游离钙离子浓度,并采用AO-EB荧光染色、流式细胞术和DNA琼脂糖凝胶电泳检测细胞凋亡。结果显示半边莲可提高HepG2细胞胞内游离钙离子浓度,与对照组相比,差异显著(P<0.01),且荧光染色、流式细胞术和DNA琼脂糖凝胶电泳的结果表明经半边莲处理过的肝癌细胞出现典型的凋亡特征。

To investigate the hepatoma cells apoptosis induced by Lobelia chinensis Lour.through the calcium pathway.The level of cytoplasmatic free Ca^(2+) was detected by fluorescent colorimetry.The apoptotic cells were detected by AO-EB double fluorescent staining,flow cytometry and DNA agarose gel electrophoresis.The results showed that Lobelia chinensis Lour.could significantly increases the level of cytoplasmatic free Ca^(2+) in HepG_(2)(P<0.01),and HepG_(2) showed typically apoptotic character.Conclusion: Lo...
