摘要
探讨大蒜油和白藜芦醇联合用药对人胃癌MGC-803细胞的抑制作用及其机制。通过MTT法检测大蒜油和白藜芦醇联合用药对胃癌细胞的抑制作用;流式细胞术检测药物作用后细胞周期改变;RT-PCR方法检测联合用药后Bcl-2、c-myc和hTERT基因mRNA表达水平。结果表明药物作用24 h和48 h后,联合用药对胃癌细胞的抑制率高于单药组,并表现明显的协同作用,可使胃癌细胞阻滞于细胞周期的G0/G1期;联合用药24 h和48 h后胃癌细胞Bcl-2、c-myc和hTERT基因mRNA表达水平比对照组明显下降。说明大蒜油和白藜芦醇联合用药能明显抑制胃癌细胞增殖并有协同作用,其机制可能与抑制胃癌细胞Bcl-2、c-myc和hTERT基因表达有关。
To explore the inhibitory action of garlic oil combined with resveratrol on human gastric cancer cell line MGC-803.The inhibition rate of cell proliferation was measured by MTT,cell cycle was analysed by flow cytometry,and the mRNA expressions of Bcl-2,c-myc and hTERT gene were determined by RT-PCR method.The results showed that garlic oil combined with resveratrol inhibited the proliferation of human gastric cancer cell markedly and their synergistic effect could be observed.The cell cycle was blocked to G...
