贵阳市区绝经后女性Ⅰ型胶原α1链Sp1结合位点基因多态性的研究

摘要 目的探讨贵阳地区汉族绝经后女性I型胶原α1链(COL1A1)第一个内含子内Sp1结合位点基因多态性与超声骨量及骨代谢状态的关系。方法采用PCR-RFLP法检测COL1A1的Sp1结合位点基因多态性:用sunlight omnisense 7000TM型超声骨密度仪测量其左侧桡骨远端、左侧中指第三指骨、右侧胫骨中段SOS;血清骨钙素(BGP)测定采用放射免疫法,血清甲状旁腺激素(PTH)及I型胶原氨基末端前肽(PINP)的测定采用酶联免疫法。结果COLlAl SPI结合位点的基因型均为SS型,未发现Ss和ss基因型。结论COLlAl基因多态性可能存在较大的种族差异,贵阳地区汉族绝经后女性COL1A1基因Spl位点多态性与骨量及骨折的关系尚不明确,该基因尚不能作为筛查贵阳地区骨质疏松高危人群的候选基因。 Objective To investigate the relationship between the Polymorphism of Sp1 combine site in Collagen type Iα1 gene and ultrasound bone mass or the condition of bone metabolism.Methods The polymorphisms of Sp1 combine site in Collagen type Iα1 gene were observed by PCR-RFLP.The multisides ultrasound bone mass were measured by quantitative ultrasound scan system(sunlight omnisense 7000^(TM),Myriad,Isereal);the blood concentration of BGP,PTH and PINP were determined by ELISA.Results Neither Ss nor ss genotypes o...
