摘要
目的:探讨神经元突起定向生长和突触形成过程的规律。方法:采用RT-PCR方法检测大鼠发育不同阶段胶质细胞源性netrin-1、nogo-A和tsp-1 mRNA的表达。结果:RT-PCR实际扩增长度与设计长度相吻合,未出现与DNA相同的杂带。内参照β-actin的PCR产物电泳条带在胚胎、新生和成年时灰度较均一。netrin-1 mRNA以胎鼠和新生大鼠表达较多,成年大鼠弱表达(P<0.01)。nogo-A在各个阶段均呈高表达,以胎鼠和新生大鼠较多,其中新生大鼠表达最多(P<0.01),成鼠相对较低(P<0.01)。tsp-1以胎鼠和新生大鼠表达较多,成年大鼠表达微弱(P<0.01)。结论:大鼠海马胚胎和新生后是突起定向生长的主要阶段,抑制突起的生长则发生在发育的各个阶段,以新生后为主,突触结构和功能的发育发生在胚胎和新生后阶段。
Aim: To investigate the rule of neurite orientating growth and synapse formation.Methods: The mRNA of netrin-1,nogo-A and thrombospondins-1(tsp-1) was detected using RT-PCR.Results: Limpid electrophoresis strips located in the contrived position,and no hetero-strips appeared.The expression of β-actin was similar in all rats.The most expression of netrin-1 was in embryonic and neogenetic rats,and the least was in adult rats(P<0.01).The expression of nogo-A was more in all period and mainly in the embryoni...
