人类脐带血间充质干细胞的培养及鉴定

摘要 目的对人类脐带血中的有核细胞进行分离培养及鉴定,观察能否得到间充质干细胞。方法抽取人类脐带静脉血,进行细胞分离和培养,并进行荧光激活细胞分析。结果脐带血中间充质干细胞与骨髓中的类似,其外表呈纤维样;流式细胞术鉴定结果表明这种细胞能表达CD29、CD44、CD90、CD95、CD105、CD166以及MHCⅠ(组织相容性抗原复合体Ⅰ),但不能表达CD14、CD34、CD40、CD45、CD80、CD86、CD117、CD152以及MHCⅡ(组织相容性抗原复合体Ⅱ);并且具有分化成骨细胞和脂肪细胞的潜能。结论可以从人类脐带血中成功分离培养得到间充质干细胞。 Purpose To cultivate and identify mesenchymal stem cells(MSCs) from human umbilical cord blood.Methods Human umbilical cord blood was drawn out,then MSCs was separated and cultivated.They were identified by FAC-Scan.Results MSCs were found in human umbilical cord blood with fibroblast-like appearing.Flow Cytometry results showed that they expressed CD29,CD44,CD90,CD95,CD105,CD166 and MHC class Ⅰ,but the expression of CD14,CD34,CD40,CD45,CD80,CD86,CD117,CD152 and MHC class Ⅱ was negative.These cells had the ...
