摘要
目的:测定并比较实验性1型糖尿病大鼠模型与正常对照组大鼠胰岛α细胞中胰高血糖素(GLC)、神经肽Y(NPY),β细胞中胰岛素蛋白表达,明确T1DM大鼠胰岛中α、β细胞的功能改变及相互关系。方法:用普通饮食/大剂量链脲佐菌素制备T1DM大鼠模型;测定两组大鼠生化代谢指标及胰岛面积;采用荧光双标及单标法分别对两组大鼠胰岛GLC、NPY,INS定位分析及α细胞中NPY,GLC,β细胞中INS定量分析。结果:实验性T1DM鼠模型的生化代谢指标与临床疾病表现一致;与正常对照组相比,T1DM组胰岛面积缩小,β细胞中INS表达减少,而α细胞中的NPY,GLC表达明显增高(P<0.01)。表达NPY及GLC的α细胞分布方式发生由正常的外周向中央分布的改变。结论:本实验证明T1DM发生时,INS表达减少与NPY,GLC表达增多同时并存,为T1DM的发病机理的研究及防治提供新思路。
