摘要 研究含有ATP结合转运子A1基因的真核表达载体pcDNA3.1/ABCA1转染真核细胞后,在细胞中的表达及对细胞抗砷性的影响。由脂质体介导将pcDNA3.1/ABCA1转染入Hela细胞中,用Western-blot方法检测ABCA1蛋白的表达,用MTT法检测48h急性砷染毒后细胞生存率的改变。Western结果显示重组质粒成功转入Hela细胞中且表达良好,MTT结果显示转染重组质粒组细胞在各浓度下生存率均较对照组高。这表明,ABCA1蛋白在真核细胞中相对高表达后增强了细胞的抗砷性。 To examine the expression of ATP-binding cassette transporter A1 in eukaryotic cells and the effect of arsenic resistance after transfect the eukaryotic expression vector.Hela cells were transfected with the recombinant plasmid and the expression of recombinant ABCA1 was examined by Western blot.The change of cell survival rate was examined by MTT after exposure in arsenic for 48 hours Western blot shows that recombinant plasmids were successfully transfected into Hela cells.MTT shows that the survival rate...
