托吡酯对戊四氮点燃大鼠海马生长相关蛋白-43 mRNA表达的影响

摘要 目的:观察托吡酯对戊四氮点燃大鼠海马结构内生长相关蛋白(GAP)-43mRNA表达的影响。方法:健康成年雄性SD大鼠40只,随机分为4组,每组10只。I组大鼠胃内注入生理盐水(10mL/kg)后1h腹腔注射10g/L戊四氮(35mg/kg);Ⅱ组和Ⅲ组大鼠腹腔注射戊四氮(剂量同I组)前1h分别按100mg/kg和40mg/kg胃内注入10g/L托吡酯生理盐水溶液;Ⅳ组大鼠按10mL/kg和3.5mL/kg分别胃内注入和腹腔注射生理盐水,间隔1h。各组选取2个研究时间点,第1个时间点为I组大鼠性发作达到Ⅲ级,第2个时间点为Ⅰ组大鼠性发作达到Ⅴ级,每个时间点各组大鼠为5只。应用RT-PCR半定量检测不同时间点各组大鼠齿状回、海马CA3区及CA1区GAP-43mRNA的表达。结果:2周和4周时I组大鼠性发作分别达到Ⅲ级和V级。GAP-43mRNART-PCR产物经溴乙啶显色后显示出256bp和385bp2个预期的条带。在同一时间点,Ⅰ组、Ⅱ组和Ⅲ组大鼠齿状回及CA3区的GAP-43mRNA表达强于Ⅳ组,差异有统计学意义(P<0.05);各组在CA1区GAP-43mRNA表达差异无统计学意义(P>0.05);Ⅱ组和Ⅲ组大鼠齿状回和CA3区的GAP-43mRNA表达水平较Ⅰ组降低,差异有统计学意义(P<0.05),而Ⅱ组和Ⅲ组大鼠之间GAP-43mRNA表达水平差异无统计学意义(P>0.05)。与2周时相比,4周时Ⅰ组大鼠GAP-43mRNA表达水平进一步增强,差异有统计学意义(P<0.05)。结论:戊四氮点燃大鼠海马GAP-43mRNA表达增强,表明突触重建参与了癫发生的病理过程;托吡酯可抑制GAP-43的表达,可能是托吡酯抗癫作用的机制之一。 Aim:To explore the expressional character of growth-associated protein-43(GAP-43)and observe the effects of topiramate(TPM)on the expression of GAP-43 in the hippocampal formation of rats kindled by pentyenetrazol(PTZ).Methods:A total of 40 healthy adult male SD rats were randomly divided into 4 groups(n=10).The rats of group I were given normal saline at 10 mL/kg by gavage and intraperitonealy injected 10 g/L PTZ(35 mg/kg)daily.The rats of groupⅡ and group Ⅲwere given 10 g/L TPM at 100 mg/kg and 40 mg/kg,r...
