提高粳稻花药小孢子离体培养反应的研究 被引量:6

摘要 以大田种植的粳稻品系为供试材料,比较了基因型、离体穗低温预处理和诱导培养基中0.5mg/L2,4-D、2,4,5-T与50g/L蔗糖、麦芽糖配合使用、渗透压以及添加0.5mg/LNAA对粳稻花药培养的影响.结果表明:基因型对培养效果有着重要的影响,敏感性与迟钝型在愈伤组织产量和绿苗数量上有较大的差异;对离体穗进行5℃8d低温预处理培养效果比5℃4d的好;诱导培养基中0.5mg/L2,4,5-T和50g/L蔗糖搭配获得了最佳的培养效果.以05P-27和05P-40为供试材料,培养其游离小孢子,获得了大量胚状体和绿苗. The japonica rice lines growing in field were used as experimental materials.The effects of genotype,low temperature pretreatment of spikes,2,4-D,2,4,5-T combined with sucrose or maltose in induction medium on in vitro culture of rice anther were investigated.The culture response of anthers was critical to the donor genotype and a large number of callus and green plantlets were obtained from high responding lines.The culture response of anther from pretreatment of spikes at 5℃ for 8 days was higher than tha...
