摘要
在模拟人体生理环境下,用荧光分光光度法及紫外-可见分光光度法研究了解热镇痛药对乙酰氨基酚(简写作PTL)与牛血清蛋白(BSA)之间的相互反应。试验结果发现:由于非辐射能量转移引起的荧光猝灭,BSA的荧光强度因与PTL相互之间的结合反应而明显降低。按Stern-Volmer方程和Lineweaver-Burk方程对所得实际数据进行了处理,求得此结合反应的静态平衡常数(KLB)为2.592×103mol.L-1(297 K),其结合位置距212位色氨酸1.94 nm。根据结合反应的热力学参数,推断PTL与BSA之间的结合力为疏水作用力。
The binding reaction of the drug,paracetanol(PTL),with bovine serum albumin(BSA) was studied by fluorescence spectrophotometry and UV-VIS spectrophotometry,in simulating physiological environment of human body.It was found that the fluorescence intensity of BSA was remarkably reduced due to fluorescence quenching through non-radiative energy transfer.Experimental data were treated by the stern-Volmer equation and the LineweaverBurk equation,and the static equilibrium constant(KLB) of the binding reaction wa...
