苦参素对肾小球系膜细胞增殖及TGF-β_1、α-SMA合成的影响 被引量:1

摘要 目的研究苦参素对肾小球系膜细胞增殖及TGF-β1、α-SMA合成的影响,阐明其对肾小球系膜细胞的作用。方法采用肾小球系膜细胞培养方法和甲基噻唑基四唑法(metyl thiazolyl tetronzolium,MTT)及免疫组化方法分别检测苦参素培养后肾小球系膜细胞增殖及TGF-β1、α-SMA合成的影响。结果一定浓度的苦参素能明显促进肾小球系膜细胞的增殖(P<0.05),抑制TGF-β1的合成(P<0.05),而对α-SMA合成无明显影响(P>0.05)。结论一定浓度的苦参素可以抑制肾小球纤维化,保护肾小球系膜细胞,而对系膜细胞的表型转化无明显作用。 Objective To investigate the effects of oxymatrine on cell proliferation and syntheses of TGF-β1 and α-SMA in human mesangial cells. Methods On the human mesangial cells cultured in vitro, MTT assay was used to assess cell proliferation, immunohistochemical staining was conducted to determine the syntheses of TGF-β1 and α-SMA under the effect of oxymatrine. Results Certain concentration of oxymatrine was found to have marked enhancing effect on cell proliferation(P<0.05), inhibitng effect on the synthesis of TGF-β1(P<0.05), but no effect on the synthesis of α-SMA (P>0.05). Conclusion Certain concentration of oxymatrine can inhibit kidney fibrosis and protect mesangial cells, but have no effect on the phenotype of mesangial cells.
