砷和香烟烟气溶液联合作用对大鼠淋巴细胞核因子κB的影响 被引量:4

Combined Effect of Arsenite and Cigarette Smoke Solution on NF-κB in Lymphocytes of Rats

摘要 目的研究亚砷酸钠和香烟烟气溶液(CSS)联合作用对核因子κB(NF-κB)的影响。方法将雄性Wistar大鼠淋巴细胞分为4组:亚砷酸钠单独作用组(亚砷酸钠:20μmol/L;CSS:0支/ml)、CSS单独作用组(亚砷酸钠:0μmol/L;CSS:0.008支/ml)、亚砷酸钠和CSS联合作用组(亚砷酸钠:20μmol/L;CSS:0.008支/ml)和对照组(均不暴露于砷酸钠和CSS),染毒后用凝胶迁移率变动分析检测NF-κB与DNA的结合水平,用免疫印迹检测其内源性抑制蛋白IκBα表达情况。结果亚砷酸钠单独作用组、CSS单独作用组的NF-κB与DNA结合水平显著高于对照组(P<0.05),而两者联合作用组的NF-κB与DNA结合水平却显著低于两个单独作用组(P<0.05)。亚砷酸钠单独作用组、CSS单独作用组IκBα的蛋白表达显著低于对照组(P<0.05),而两者联合作用组IκBα的蛋白表达却显著高于两个单独作用组(P<0.05)。结论亚砷酸钠和CSS单独作用能够增高NF-κB与DNA结合水平,而两者联合作用却使这种增高作用消失。 Objective To investigate the effect of sodium arsenite combined with cigarette smoke solution on NF-κB in rat lymphocytes. Methods Rat lymphocytes were divided into 4 groups: the arsenite treatment group, the CSS treatment group, the arsenite and CSS treatment group, and the control group. Electrophoretic mobility shift assays was used to detect levels of NF-κB DNA binding. Western blot was used to detect protein expression of IκBα. Results Levels of NF-κB DNA binding in the CSS treatment group and the arsenite treatment group were significantly increased (P<0.05). Levels of NF-κB DNA binding in the arsenite and CSS treatment group were significantly lower than those in the CSS treatment group and the arsenite treatment group (P<0.05). Levels of IκBα protein expression in the CSS treatment group and the arsenite treatment group were significantly decreased (P<0.05). Levels of IκBα protein expression in the arsenite and CSS treatment group were significantly higher than those in the CSS treatment group and the arsenite treatment group (P<0.05). Conclusion The arsenite treatment and the CSS treatment alone can lead to significant increase in levels of NF-κB DNA binding, but this single effect is not seen when the combined treatment is used.
