摘要
目的:观察环氧化酶-2(Cyclooxygenase-2,COX-2)选择性抑制剂塞来西布对化学致癌剂7,12-二甲基苯蒽(7,12-dimethybenzanthracene,DMBA)化学诱发的大鼠乳腺癌形成的影响。方法:DMBA油剂灌胃复制大鼠乳腺癌模型,每组30只大鼠,分为:单纯诱癌组、三苯氧胺组、塞来西布组,观察各组肿瘤发生率、潜伏期、肿瘤数目及体积的差异。结果:三苯氧胺组的肿瘤发生率、肿瘤发生时间、肿瘤数目、肿瘤总体积分别为48.15%(13/27)、97.54±1.85d、1.77±0.73个、1.78±0.71cm3;塞来西布组的分别为50.00%(14/28)、96.79±2.89d、1.71±0.61个、2.05±1.04cm3;明显低于单纯诱癌组85.71%(24/28)、89.50±5.99d、3.50±1.62个、6.42±3.96cm3,差异有显著性意义(P<0.05)。结论:塞来西布能抑制DMBA诱发的大鼠乳腺癌的发生、发展。
Objective: To evaluate the chemopreventive effect of COX-2 inhibitors on DMBA- chemically induced breast cancer. Methods: 7,12 - dimethylbenz anthracene was irrigated into the stomach of SD female rats to build breast cancer model, and the rats were divided into three groups: control group,tamoxifen group and celecoxib group. The lantency period, number and volume of breast cancer were detected and analysed. Results: The incidence of tumor occurrence, the lantency period, number and volume of tamoxifen group were 48.15%(13/27), 97.54±1.85day, 1.77±0.73, 1.78±0.71cm3 respectively; celecoxib group were 50.00%(14/28), 96.79±2.89day, 1.71±0.61, 2.05±1.04cm3 respectively; the incidence of tumor occurrence, lantency period, number, diameter and volume of chemopreventive group were lower than those of control group, and there was significant significance(P<0.05). Conclusion: Celecoxib is an effective agent method to prevent the occurrence of DMBA- chemically induced rat breast cancer.
