流式细胞术在子宫内膜疾病DNA分析中的应用

摘要 目的:探讨流式细胞术在子宫内膜疾病DNA分析中的临床应用价值。方法:应用流式细胞术对不同病变程度的石蜡包埋及新鲜的子宫内膜细胞DNA倍体含量和细胞周期时相进行分析。结果:子宫内膜增殖症组的异倍体率与正常对照组间的差异无统计学意义(P>0.05);与高分化子宫内膜癌组和低分化子宫内膜癌组间的差异均有统计学意义(P<0.05);DI、SPF介于正常对照组与高分化子宫内膜癌组之间,且与各组之间的差异均有统计学意义(P<0.05),各组细胞的DI、SPF值彼此有交叉现象;各组石蜡包埋的子宫内膜组织与新鲜子宫内膜组织之间的异倍体率和SPF差异均无统计学意义(P>0.05)。结论:流式细胞术DNA测定在子宫内膜病变诊断和评估预后中具有重要价值,而新鲜的子宫内膜流式细胞术分析操作更加简便、快速。 Objective:To investigate the clinical value of DNA assay in patients with endometrial disease by flow cytometry(FCM).Methods:The contents of DNA and cell cycle phase of different pathological changes of paraffin-embedded specimens and fresh endometrial cells were determined by flow cytometry.Results:The difference of aneuploidy between the endometrial proliferation group and the normal control was of no statistical significance(P >0.05).There was statistical significance between the endometrial proliferation group and the high polarization of endometrial cancer and between the endometrium proliferation group and the low polarization of endometrial cancer(P <0.05).DNA index(DI)and S-phase fraction(SPF)of endometrial proliferation group were between the normal control and the high polarization of endometrial cancer,and the differences between each group showed statistical significance.There were cross phenomena of DI and SPF between each group.The differences of aneuploidy and DI and SPF between paraffin-embedded specimens and fresh endometrial cells from each group thowed no statistical significance.Conculusion:FCM of DNA assay has important value in the diagnosis of endometrial disease and the evaluation of prognosis.FCM of fresh endometrium analysis was simple,fast and so it easy to use in clinic.
