摘要
目的观察巨噬细胞在刀豆蛋白A(Con A)诱导的免疫性肝脏损伤模型中的作用。方法制备骨膦脂质体,通过尾静脉注射到小鼠体内诱导巨噬细胞凋亡,再用Con A诱导小鼠肝脏损伤,利用酶法检测小鼠血浆转氨酶水平、RT-PCR法检测肝脏组织中TNF-αmRNA表达水平和H-E法分析肝脏组织病理学变化。结果骨膦脂质体体外可以诱导巨噬细胞凋亡,体内注射可以降低Con A诱导小鼠血浆转氨酶升高、下调肝脏组织中TNF-αmRNA表达以及改善小鼠肝脏组织病理损伤程度。结论巨噬细胞在Con A诱导的肝脏损伤中是重要效应细胞之一。
OBJECTIVE To investigate the role of macrophages in the model of hepatic injury mediated by concanavalin A.METHODS Cl2mbp-liposome is prepared by thin-film method;Cl2mbp-liposome induce apoptosis of macrophages in mice by tail intravenous administration,and then concanavalin A induced hepatic injury in mice by tail intravenous administration.Blood density of ALT is examined by ferment method;TNF-αmRNA is examined by RT-PCR method;The pathological change of hepatic tissue is examined by H-E method. RESULTS Cl2mbp-liposome induces the apoptosis of macrophages in vitro;Concanavalin A can improve the density of blood ALT and reduce the density of TNF-αmRNA,and restore the damage of hepatic tissue.CONCLUSION Macrophages play a important role in the hepatic injury induced by concanavalin A.
