摘要
目的检测兔真菌性角膜炎中基质金属蛋白酶-1(MMP-1)的表达,探讨MMP-1在真菌性角膜炎发病机制中的作用及其选择性抑制剂GM6001对其表达的影响。方法75只新西兰大白兔随机分为3组:对照组、真菌性角膜炎组和GM6001治疗组。分别于接种真菌后12h,24h,4d,7d,14d,21d,28d用逆转录聚合酶链反应(RT-PCR)方法检测角膜组织中MMP-1 mRNA的表达。结果对照组及12h时相兔角膜组织中,未检测到MMP-1mRNA表达,24h时相检测到MMP-1 mRNA表达,与对照组比较,差异有显著性(P<0.05)。随时间延长,MMP-1mRNA表达量逐渐升高,并持续至28d。自24h时相起,GM6001治疗组各时相兔角膜组织中MMP-1均有表达,但明显低于角膜炎组,差别有统计学意义(P<0.05)。结论MMP-1与真菌性角膜炎组织溶解密切相关,GM6001对MMP-1的表达和活性有明显的抑制作用,进而抑制角膜组织的溶解。
Objective To detect the expression of Matrix Metalloproteinases-1(MMP-1) in rabbit cornea tissue of fungal keratitis(FK),to explore its role in FK and its selective inhibitor GM6001 effect.Methods Seventy-five New Zealand rabbits were randomly divided three groups:control group,FK group and GM6001 treatment group.Through Reverse Transcription-polymerase chain reaction(RT-PCR) method to detect the expression of MMP-1 at 12h,24h,4d,7d,14d,21d and 28d after inoculated fungus in cornea tissue.Results In control group and 12h rabbit cornea tissue of fungal keratitis,can not detect the expression of MMP-1 mRNA.But in 24h fungal keratitis group can detect the expression,and have significant difference with control group(P<0.05).With the time gone,the expression of MMP-1 mRNA was increasing until to 28d.In GM6001 treatment group all can detect the expression of MMP-1 mRNA from 24h,but significantly lower compared with the fungal keratitis(P<0.05).Conclusions MMP1 is closely related with the histolysis of fungal keratitis.GM6001 has significantly inhibition effect on MMP-1 expression and activity,and further to inhibit the cornea histolysis.
