摘要
用RAPD分子标记对4种耐寒植物进行了基因组DNA遗传多样性分析,从35个随机引物中筛选出了扩增谱带清晰并呈现多态性的20个随机引物,每个引物能产生1-10条RAPD片段,多态性条带占67.61%。利用UPGMA聚类法对4种卫矛属植物的176条谱带进行聚类分析,结果表明,北海道黄杨和扶芳藤聚类成一组,大叶黄杨和金心黄杨聚类成另一组,可见,RAPD技术对卫矛属植物的分类鉴定和遗传多样性评价是有效可行的。
Random amplified polymorphic DNA(RAPD) technique was used in analyzing the biological genetic diversity of 4 cold-resistant plants.20 clearly expanded bands with kinds of shapes primers from 35 arbitrary primers were used to amplify 176 DNA bands,of which polymorphic RAPD loci covered an average of 67.61%.The UPGMA cluster analysis showed that the 4 cold-resistant plants could be divided into 2 main groups.Group one consisted of Euonymus japonicus Thunb and Euonymus fortunei.Group two consisted of cv.aureo-variegatus and Euonymus japonicus.RAPD is valuable in studying on biological genetic diversity of Celastraceae.
