摘要
本文以我国鲤鱼主要养殖品种——建鲤为研究对象,运用AFLP技术分析比较雌雄鲤鱼之间的差异。共使用64对引物组合对雌雄个体进行扩增,从中筛选出9对引物,电泳图谱条带丰富,多态性位点比例高,每对引物检测出的位点数从104-134个不等,平均121.9个,多态位点比例占69.0%-92.3%;9对选择性扩增引物共扩增出1097个片段,其中264个片段(24.1%)为所有雌雄个体共有条带,其余833个(75.9%)为多态性片段;统计分析,共发现18条带在不同性别中出现的比例差异显著。
Jian carp(Cyprinus carpio var.jian),with excellent economic traits,is an extensively cultured variety of common carp in China.As a result,Jian carp was chosen as experimental material in this paper.AFLP technique was used to screen the sex-related DNA marker through analyzing the DNA differences between female and male of Jian carp.9 pairs of AFLP primer were selected from 64 primer combinations to detect the genome DNA of the female and male individuals by AFLP analysis.Totally 1097 bands were obtained with AFLP analysis,of which 264 bands shared by female and male individuals(the proportion was 24.1%),and the rest 833 bands were polymorphic fragments(the proportion was 75.9%).Big differences were found in the frequency of some bands between female and male population,which were probably related to sex.Based on the result of statistics,the ratio between female group and male one of 831 amplified loci displayed the difference in gene frequency.18 of them show apparently different distribution between female group and male one.The above data were also helpful for understanding the genetic background,establishing the germplasm standards and genetic improvement of Jian carp.
