摘要
目的探讨尿道下裂患者5a-还原酶2(SRD5A2)表达和睾酮值异常。方法选择20例轻至中度尿道下裂患儿为病例组,另选20例尿道外口正常儿童作为对照组。应用RT-PCR方法对所有样本包皮组织中SRD5A2-mRNA的表达进行检测,并结合计算机图像分析病例组和对照组的SRD5A2的含量进行比较;同时抽血查2组睾酮值。结果所有样本中均有SRD5A2表达,尿道下裂患者和尿道外口正常儿童包皮组织中SRD5A2-mRNA表达水平相比差异没有显著性(P>0.05)。两组睾酮值均在正常范围,相比差异没有显著性(P>0.05)。结论5a-还原酶2(SRD5A2)可能与轻至中度先天性尿道下裂的发生无关。轻至中度先天性尿道下裂睾酮合成并不受影响。
[Objective] To investigate the expression of Steroid-5α Reductase type2(SRD5A2)and determination of testosterone in children with hypospadias.[Methods] The expression of SRA5A2 was detect in the prepuce tissue from 20 boys with hypospadias and 20 controls using reverse transcription polymerase chain reaction(RT-PCR).The SRD5A2 levels were analysed by computer digital image analysis.At the same time,we determine the testosterone.[Results] SRD5A2 expression was present in the foreskin from both groups.The levels of foreskin SRD5A2 had no significant changes in the patients with hypospadias as that of control group(P >0.05).The value of testosterone was normal and also had no significant changes(P >0.05).[Conclusions] The expression of SRD5A2 may be unrelated to the incidence of the isolated hypospadias.The composition of testosterone don't influence the isolated hypospadias.
