IL-1β影响大鼠星形胶质细胞中SSeCKS表达的研究

摘要 目的:研究Src抑制的蛋白激酶C的底物(SSeCKS)在IL-1β激活的星形胶质细胞中的表达。方法:以炎性因子IL-1β刺激大鼠体外培养的星形胶质细胞,观察细胞中SSeCKS的表达及其细胞内的亚定位的改变。结果:IL-1β可上调大鼠原代培养星形胶质细胞中SSeCKS的表达,诱导其丝氨酸磷酸化。在亚细胞水平,IL-1β还能引起SSeCKS向核周、细胞星状突起聚集,而蛋白激酶C(PKC)抑制剂Ro-31-8220能抑制IL-1β对细胞中SSeCKS亚细胞定位及磷酸化水平的影响。结论:在IL-1β的影响下,大鼠星形胶质细胞中SSeCKS的表达增加,磷酸化水平增强,细胞内定位发生改变,这种改变与PKC的功能相关,提示SSeCKS可能作为PKC的下游分子参与到炎性因子引起的星形胶质细胞活化的过程中。 Objective:To investigate the effect of Interleukin-1β(IL-1β) on SSeCKS expression in rat primary astrocytes.Methods:Purified astrocytes from the cortical cortex of newborn rats were incubated with IL-1β.Western blot,Realtime PCR and immunofluorescent staining with confocal laser-scanning microscope were used to examine expression and subcellular localization of SSeCKS.Results:IL-1β enhanced the expression and serine phosphorylation of SSeCKS in the cultured rat astrocytes.In microscope, SSeCKS translocated to the stellate processes and perinuclear sides while the PKC inhibitor RO-31-8220 could partly reverse ssecks eranscocation.Conclusion:IL-1β induces the expression and serine phosphorylation of SSeCKS,coincident with its translocation to the stellate processes and perinuclear sides .These effects is PKC dependent. Suggesting a role of SSeCKS as a downstream mediator of PKC in the process of IL-1β induced astrocytes activiation.
