重组人生长激素联合奈达铂在体外对人食管癌细胞株EC109作用的实验研究 被引量:6

摘要 目的 研究重组人生长激素(rhGH)在体外对人食管癌细胞生长的影响.方法 实验分为对照组、rhGH组、奈达铂(nedaplatin,NDP)组和rhGH+ NDP 组共4组,利用体外细胞培养、MTT比色法、流式细胞术及免疫细胞化学等方法,测定不同浓度的rhGH对人食管癌细胞株EC109细胞抑制率、细胞周期、增殖指数(PI)及核增殖抗原Ki-67表达的影响.结果 rhGH在体外不能明显促进EC109细胞分裂增殖,对照组与rhGH组、rhGH+ NDP组与NDP组比较细胞抑制率、PI、Ki-67表达强度差异均无统计学意义(均P>0.05);rhGH各亚组间比较差异亦无统计学意义(均P>0.05);NDP组、rhGH+NDP组与rhGH组、对照组比较细胞抑制率增加,阻滞于G0~G1期的细胞数增加,S期细胞明显减少,PI明显降低,Ki-67表达强度明显下降(均P<0.05).结论 重组人生长激素在体外不能明显促进食管癌EC109细胞生长. Objective To investigate the effects of recombinant human growth hormone(rhGH)on growth of human esophageal carcinoma cell line EC109 in vitro.Methods Four groups were assigned:control group,rhGH group,NDP group and rhGH + NDP group.Effects of different concentrations of rhGH on cell inhibitory rate,cell cycle proliferation index(PI)and ki-67 express intensity of esophageal carcinoma cell line EC109 were studied by cell culture,MTT assay,flow cytometry and immunocytochemistry.Results In vitro the distinctly accelerating effects of rhGH on multiplication of EC109 cell line were not found.There was no statistically significant difference of cell inhibitory rate,PI and ki-67 express intensity between rhGH group and control group or between rhGH + NDP group and NDP group(all P>0.05).Cell inhibitory rate and the cells arrested in G0/G1 phase were obviously increased,while the cells in S phase and PI distinctly decreased in NDP group or rhGH + NDP group as compared with control group or rhGH group,also ki-67 express intensity distinctly decreased(P<0.05).Conclusion In vitro rhGH does not accelerate the multiplication of human esophageal carcinoma cell line EC109.
