Synphilin-1寡核苷酸对帕金森病caspase-8表达影响的实验研究 被引量：2

摘要 目的利用动物模型探讨Synphilin-1对帕金森病(PD)caspase-8蛋白表达的影响。方法50只SD大鼠随机分成5组,每组10只,第一组立体定向将6-羟多巴注入右侧黑质制作PD模型,术后每周用阿朴吗啡诱发旋转30 min,至第四周旋转大于6次/min者定为成功的PD模型,第二至第四组利用立体定向将Synphilin-1反义、同义、错义寡核苷酸分别注入到各组大鼠的右侧黑质,然后用与第一组相同的方法制作PD模型,第五组注入生理盐水作为对照组,取鼠的术侧中脑黑质利用免疫组化(IH)检测Synphilin-1,利用IH、原位杂交(ISH)检测caspase-8的表达。结果IH检测Synphilin-1蛋白,与正常对照组比较,PD组显著增加;IH、ISH检测caspase-8蛋白和mRNA表达,与正常对照组比较,PD组显著增多,与PD组比较,反义组表达显著减少。结论Synphilin-1在PD的重要发病机制之一可能通过影响caspase-8的表达来实现。 Objective: To investigate the relation between Synphilin-1 and caspase-8 of Parkinson disease.Methods: Fifty healthy male SpragueDawley rats were divided into five groups.The first group of ten rats were prepared to be hemiparkinsonian models induced by unilateral stereotaxic infusion of 6-hydroxydopamine(6-OHDA) in right substantia nigra of midbrain.Every week after operation,The circles the rats run were observed in 30 minute after injecting apomorphine by peritoneal cavity.If the rat ran more than 6 circ...
