氨溴索对百草枯中毒性急性肺损伤大鼠肺组织HO-1表达的影响 被引量:6

摘要 目的探讨氨溴索治疗百草枯(PQ)中毒性急性肺损伤的效果及机制。方法将120只大鼠随机分为4组,正常对照组(C组)、氨溴索对照组(AC组)各12只,均腹腔注射生理盐水1.25ml/kg,AC组再腹腔注射氨溴索35mg/kg;PQ中毒组(PQ组)及氨溴索治疗组(AT组)各48只,均腹腔注入2%PQ液25mg/kg制造中毒模型;AT组造模后腹腔注射氨溴索35mg/kg,1次/d。PQ组、AT组于腹腔注射PQ后1、3、5d各处死16只,C组、AC组各处死4只。取肺组织HE染色评价肺组织损伤情况;检测血清中IL-1β水平;采用RT-PCR法检测肺组织HO-1 mRNA表达;采用Western blot方法分析肺组织HO-1蛋白表达。结果C组、AC组肺组织结构基本正常,PQ组肺组织损伤较C组明显加重,IL-1β的表达水平明显升高(P<0.01),肺组织的HO-1 mRNA和蛋白表达水平亦升高(P均<0.05)。而AT组肺组织损伤较PQ组有所减轻,IL-1β的表达水平亦降低(P<0.01),肺组织的HO-1 mRNA和蛋白表达水平皆高于PQ组(P均<0.01)。结论氨溴索可减轻大鼠PQ中毒所致的肺损伤,其机制可能与降低IL-1β表达及升高肺组织中HO-1基因和蛋白表达有关。 Objective To explore the effect and mechanism that ambroxol treats the acute lung injury of rats associated with paraquat poisoning.Methods 120 Rats were randomly divided into four groups:C group(control group,NS 1.25 ml·kg-1,ip),AC group(ambroxol control group,ambroxol 35 mg·kg-1,ip and after NS 1.25 ml·kg-1,ip),PQ group(paraquat group,2% paraquat 25 mg·kg-1,ip),AT group(ambroxol treatment group,ambroxol 35 mg·kg-1 after 2% paraquat,ip).Four groups were divided into 1 d,3 d,5 d subgroups further.The injury...
