贞清方对2型糖尿病大鼠肾组织基质金属蛋白酶-9和基质金属蛋白酶组织抑制物-1表达的调节作用 被引量:9

摘要 目的探讨中药贞清方(含女贞子、地龙等中药)对2型糖尿病大鼠肾脏结构及肾组织基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶组织抑制物-1(TIMP-1)表达的影响。方法采用高糖高脂饲料喂养加小剂量链脲佐菌素(streptozotocin,STZ)注射的方法建立2型糖尿病模型。将成模大鼠随机分成模型组,贞清方高、低剂量组,依那普利组,并设立正常对照组,治疗8周。用免疫组化法检测MMP-9、TIMP-1以及纤维连结蛋白(FN)在肾脏的表达,光镜观察肾小球和肾小管形态。结果糖尿病模型组较对照组TIMP-1、FN表达增强,MMP-9表达减弱;治疗组TIMP-1、FN表达较模型组减弱,MMP-9表达增强。其中贞清方高剂量组作用最明显。形态学观察显示治疗组肾小球和肾小管病变比模型组减轻。结论贞清方可能通过影响糖尿病大鼠肾脏MMP-9、TIMP-1的表达而发挥其对糖尿病大鼠肾脏结构的保护作用。 Objective To explore the effect of Zhenqing Recipe(ZQR)on renal structure and expressions of matrix metalloproteinase-9(MMP-9),tissue inhibitor of matrix metalloproteinase-1(TIMP-1)in experimental type 2 diabetic rats.Methods The rat models of type 2 diabetic were set up by intraperitoneally giving small-dose streptozotocin(STZ)after fed with high carbohydrate and high fat diets for one month.The model rats were ran- domly divided into the model group,the high and low dose ZQR-treated groups,and the enalapr...
