摘要
目的观察抗脑血栓片对缺血性脑损伤的保护作用,并初步探讨其可能的作用机制。方法大鼠灌胃给予抗脑血栓片,连续14天,每天2次。采用大鼠大脑中动脉栓塞(MCAO),制作急性局灶性脑梗塞动物模型,观察缺血24h后对脑梗塞体积和神经行为学变化的影响,以及对大鼠体外血栓形成、血小板聚集、血液黏度等指标的影响。结果抗脑血栓片可依剂量性减轻大鼠局灶性脑缺血所致的脑梗塞体积以及改善由脑缺血引起的神经症状。大、中、小剂量组脑梗塞体积分别为(11.18±3.35)%、(14.60±7.00)%和(15.37±7.21)%,与模型组(27.51±4.71)%比较,各组依次分别下降59.36%、46.93%和44.13%(P<0.01),各剂量组对体外血栓的干、湿重量均亦可明显减轻,与模型组比较,差异有显著性(P<0.05,P<0.01),可显著抑制ADP诱导的血小板聚集,降低全血黏度,但对血浆黏度、红细胞压积无明显影响。结论抗脑血栓片对脑缺血有明显的保护作用,其作用可能与它改善血液黏度、抑制血小板聚集有关,对其确切作用机制,有待深入研究。
