摘要
【目的】研究犬枪弹伤后脑组织中热休克蛋白70(Heat Shock Protein70,HSP70)和bcl-2的变化规律。【方法】24只杂种犬,随机分为对照组,损伤组。采用德国小口径步枪子弹致犬颅脑额叶切线伤(TangentBrain Injury,TBI)。用免疫组织化学方法检测伤后2、6、12、24和48 h各不同时期弹道挫伤区、震荡区及脑干神经元中HSP70和bcl-2蛋白的表达。【结果】对照组脑神经元中HSP70和bcl-2蛋白表达弱,枪弹伤组挫伤区、震荡区、海马及脑干神经元中HSP70蛋白2 h开始增加(P<0.05),6 h时明显增加,12 h达高峰(P<0.01),24 h开始下降。bcl-2伤后2 h开始增加(P<0.05),24 h达高峰(P<0.01),48 h开始下降。【结论】HSP70和bcl-2蛋白在弹道挫伤区、震荡区、海马及脑干神经元均有表达,伤后两者在脑组织中表达分布范围不一致,HSP70先于bcl-2表达,可能共同参与细胞抗凋亡过程,相互间也可能存在着一定的调控作用。
【Objective】To investigate the expression and changes of heat shock protein(HSP70) and bcl2 in brain following craniocerebral gunshot wound in canine.【Methods】[WTBZ]Twenty-four canines were divided into control and injury group.The canine model of tangent brain injury(TBI) was established by German-made bullets of small-caliber rifles.HSP70 and bcl-2 positive neurons were determined by immunohistochemical method in the contusion trajectory area,concussion trajectory area,hippocampus and brain stem at 2,6,12,24 and 48 h after trauma.【Results】Weak HSP70 and bcl-2 were detected in the control group.In the injury group,positive expression of heat shock protein in neurons in the contusion trajectory area,concussion trajectory area,hippocampus and brain stem after craniocerebral gunshot wound increased at 2 h,reached the peak at 12 h,and began to decrease at 24 h.The bcl-2 positive expression in the above mentioned areas after craniocerebral gunshot wound increased at 2 h,reached the peak at 24 h and began to decrease at 48 h.【Conclusion】Induction and expression of HSP70 and bcl-2 is not similiar in the contusion area,concussion trajectory area,hippocampus and brain stem are the reaction after brain gunshot wound.The pattern and extent of expression of HSP70 and bcl-2 is not similiar in different areas after brain gunshot wound,and the induction of bcl-2 appears to be later as compared with that of HSP70.HSP70 and bcl-2 may be associate with the process of anti-apoptosis,and have regulation function each other.
