摘要
目的:研究氧自由基在体外对胃癌细胞株SGC-7901表达IL-8的影响。方法:用不同浓度的双氧水体外刺激SGC-7901,用酶联免疫吸附试验测定培养液上清中的IL-8分泌情况,用RT-PCR测定IL-8及NF-κΒ的表达,以不同浓度的抗氧化剂U74006F预处理培养的细胞,再用双氧水刺激SGC-7901,再行测定上述指标。结果:SGC-7901在所用H2O2浓度及U74006F治疗剂量下其生长无明显影响,活性氧能刺激SGC-7901IL-8mRNA表达及蛋白分泌并与H2O2浓度相关,U74006F能有效抑制H2O2对SGC-7901的刺激作用。结论:体外研究表明,胃癌细胞株SGC-7901IL-8表达受到氧自由基的调控,氧自由基可能通过激活NF-κΒ而起作用,抗氧化剂U74006F对控制IL-8的表达有一定作用,抗氧化治疗有望成为胃癌综合治疗的一部分。
Objective To study the effects of oxygen free radical on gastric cancer cell line SGC-7901 in vitro. Methods After various concentration of H_2O_2 stimulating SGC-7901 in vitro,IL-8 in cell culture supernatent with enzyme linked immunosobent assay were investigated,Expression of IL-8 mRNA and nuclear factor-Kappa B were measured after various concentrations of antioxidant U74006F coculture with SGC-7901 by RT-PCR,then H_2O_2 stimulated cells again and the same markers were measured. Results The growth and its viability of SGC-7901 cultured in vitro were affected with various therapcutic doses of H_2O_2 and U74006F.Reactive oxygen free radical could stimulate SGC-7901 to express IL-8 mRNA and protein secretion and the expression of IL-8 up-regulated with the increasing H_2O_2 concentration.The expresson of NF-κΒ mRNA under the same condition is similar to IL-8,antioxidant could suppress effectively the effects of H_2O_2 stimulating on SGC-7901. Conclusion Oxygen free radical can regulate the expression of interleukin-8,oxygen free radical can activates NF-κΒ,so.that to affect IL-8.Antioxidant U74006F has some effects to control the expression of IL-8.Antioxidant therapy maybe become an important part of comprehensive therapy in gastric neoplasm.
