小鼠日本血吸虫病肝纤维化肝脏CTGFmRNA的表达意义

摘要 目的研究结缔组织生长因子(connective tissue growth factor,CTGF)mRNA在日本血吸虫病肝纤维化小鼠肝脏中的表达状况及可能的意义。方法昆明小鼠感染尾蚴复制小鼠日本血吸虫病肝纤维化模型,模型组分别在6、10、14、18周杀鼠,对照组在6周时予吡喹酮治疗后10、14、18周杀鼠;HE染色观察小鼠肝脏病理改变,RT-PCR检测小鼠肝脏CTGFmRNA表达,免疫组化方法观察各阶段肝脏α-SMA^+细胞的表达。结果模型组10周时形成典型的肝纤维化,肝纤维化不断进展。模型组10周时小鼠肝脏CTGFmRNA表达和α-SMA^+细胞表达均达高峰,10周、14周、18周时与对照组相比均有统计学差异(P<0.05);半定量结果显示CTGFmRNA表达与α-SMA^+细胞表达呈直线相关性。结论CTGFmRNA表达增高与小鼠日本血吸虫病肝纤维化形成有密切关系;CTGF可能通过作用于活化的肝星状细胞来参与小鼠日本血吸虫病肝纤维化的进展。 Objective The expression of connective tissue growth factor (CTGF)mRNA in murine schistosomiasis liver fibrosis models and its possible significance were studied.Methods The mice infected with schistosoma japonicum were randomly divided into the model groups and the control groups and subsequently sacrificed at the 6^(th),10^(th),14^(th),and 18^(th) weeks respectively.Histological changes of murine liver were observed by HE staining.The hepatic expression of CTGF mR- NA was detected by reverse transcriptase polymerase chain reaction.Theα—SMA^+ positive cells were observed by immuno- histochemistical method.Results Murine schistosomiasis liver fibrosis models were successfully established.The expres- sions of CTGF mRNA andα—SMA^+ positive cells in the model groups peaked at the 10^(th) week and then declined.The levels of CTGF mRNA andα—SMA^+ positive cells showed significant differences at the 10^(th),14^(th),and 18^(th) weeks when compared with those in the control groups.Correlative analysis showed that there existed positive correlation between the expressions ofα—SMA^+ positive cells and CTGF mRNA.Conclusions The expression of CTGF mRNA is correlated to the development of,the murine schistosomiasis liver fibrosis;CTGF may function its biogical effects by its action on HSCs.
