摘要
应用蛋白磷酸化流式细胞分析技术研究系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMC)STAT的活化,探讨SLE患者是否存在胞内信号传递网络的异常。采用蛋白磷酸化流式细胞分析技术测定28例SLE患者PBMC基础水平以及多种细胞因子刺激后胞内pSTAT1、pSTAT3、pSTAT5的水平,以18例正常人为对照。结果:基础水平,SLE患者T淋巴细胞pSTAT5表达百分率显著高于正常对照组(P<0.01);SLE患者T淋巴细胞可能存在STAT5的持续活化;SLE患者对IL-6、IL-10反应异常。SLE患者存在胞内信号蛋白STAT的活化异常,并具有一定的特征性。
