摘要 应用蛋白磷酸化流式细胞分析技术研究系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMC)STAT的活化,探讨SLE患者是否存在胞内信号传递网络的异常。采用蛋白磷酸化流式细胞分析技术测定28例SLE患者PBMC基础水平以及多种细胞因子刺激后胞内pSTAT1、pSTAT3、pSTAT5的水平,以18例正常人为对照。结果:基础水平,SLE患者T淋巴细胞pSTAT5表达百分率显著高于正常对照组(P<0.01);SLE患者T淋巴细胞可能存在STAT5的持续活化;SLE患者对IL-6、IL-10反应异常。SLE患者存在胞内信号蛋白STAT的活化异常,并具有一定的特征性。 To investigate the activation of STAT in PBMC of patients with SLE and in order to test if abnormal intracellular signaling networks exist in SLE patients,the basic expressions levels of pSTAT1,pSTAT3 and pSTAT5 as well as those levels after stimulation with several cytokines in PBMC of 28 SLE patients and 18 healthy controls were assayed by phospho-specific flow cytometry.It was found that T lymphocytes of patients with SLE exhibited sustained activation of STAT5 and the positive percentage of pSTAT5 in SL...
