摘要
目的:建立猕猴骨髓基质细胞(bone marrow stromal cells,BMSCs)的分离、培养及鉴定的方法。方法:体外无菌抽取猕猴骨髓,采用差速贴壁的方法分离、培养猕猴骨髓基质细胞,通过相差显微镜观察其形态特点,并用流式细胞仪鉴定其表面抗原分子。结果:体外分离培养BMSCs以宽大扁平的梭形细胞为主,流式细胞术分析BMSCs免疫表型显示BMSCs CD73、CD105、GD29表达阳性,CD34、CD45、CD11b表达阴性。结论:分离培养的BMSCs具有间充质细胞的特性,可为组织工程化神经提供种子细胞。
Objective:To establish the methods of isolation,cultivation and identification of bone marrow stromal cells (BM- SCs) of rhesus monkeys.Methods:BMSCs were isolated from other cells by their strong adherence to plastic.The morphologic characters of BMSCs were observed with phase contrast microscope.Flow cytometry was employed to detect BMSCs surface antigens:CD73,CD29,CD105,CD11b,CD34 CD45.Results:Most of these monkey-derived BMSCs were big ancipita spindle cells.BMSCs were immunopositive for CD73、CD105 and ...
