摘要
通过抽蛋清、延长空气与胚胎接触时间的方法,就空气对 X 期鸡胚原壳发育的影响进行了研究。种蛋开孔0.5 cm,用蛋壳皿立即封口后进行孵化,X 期鸡胚和3日龄鸡胚的孵化率呈现出极显著的差异(P<0.01),X 期鸡胚的孵化率为0,而3日龄鸡胚的孵化率高达80.0%。种蛋抽取1.0 mL 蛋清后,X 期鸡胚和3日龄鸡胚的孵化率也呈现出极显著的差异(P<0.01),X 期鸡胚的孵化率为31.3%,3日龄鸡胚的孵化率为81.3%。开孔后在空气中暴露2h,然后封口孵化,结果孵化鸡胚的死亡明显加快(P<0.01),7日龄鸡胚的存活率立即封口组为51.6%,2h 后封口组为25%。上述实验表明,开孔与空气接触、抽蛋清使蛋内进入空气或延长空气与 X 期鸡胚接触的时间都可使鸡胚在孵化过程中的死亡加快,从鸡胚死亡的幅度来看,空气可能是影响鸡胚体外培养的最重要因素。
The effect of air on in vitro culture of X-stage chicken embryos in primary eggshells,by taking 1.0 mL of egg white with a springe and delaying sealing of the hole after 2 h was studied.The results showed that hatching rate of 3 days chicken embryos(80.0%)is better(P<0.01)than that of X-stage chicken embryos,that hatching rate of X-stage chicken embryos taken 1.0 mL of egg white (31.3%)significantly decreased(P<0.01)than that of 3 days chicken embryos(81.3%),and that live rate of chicken embryos sealed at 2h after drilling the hole on 7th days of hatching(25.0%) evidently fall down(P<0.01)than those of chicken embryos sealed immediately(51.6%).The experiment indicated that chicken embryos exposed to air by making a hole on eggshell for less than 10 rain and 2 h,and 1.0 mL of air entered by taking 1.0 mL of egg white died more rapidly.It is suggested that the air may be the most important factor on in vitro culture of X stage chichen embryos.
