摘要
目的:通过转染能特异性沉默血管内皮生长因子(VEGF)表达的小干扰RNA(siRNA),观察能否抑制人舌癌Tca8113细胞株的增殖。方法:将自行构建的两对表达siRNA的重组质粒(PU-VEGF-siRNA1,PU-VEGF-siRNA2)转染到舌癌Tca8113细胞株中,以空载体组转染为实验对照组,以正常生长的舌癌细胞做空白对照组。实验组和实验对照组经G418筛选后,与空白对照组以逆转录聚合酶链反应(RT-PCR)检测VEGF mRNA的表达,流式细胞技术分析细胞凋亡,MTT比色法检测细胞的生长抑制率。结果:与实验对照组相比,PU-VEGF-siRNA1,PU-VEGF-siRNA2重组体显著降低舌癌细胞VEGF mRNA的表达(P<0.01),MTT的结果显示分别在24、48、72、96h对细胞增殖的抑制率增加(P<0.05),细胞凋亡率也明显增高(P<0.01);而空载体转染组与空白对照组相比,对Tca8113细胞株增殖无抑制作用,对细胞凋亡变化无明显影响(P>0.05)。结论:PU-VEGF-siRNA在细胞内表达的siRNA不仅能显著降低VEGF表达,而且能有效抑制人舌癌细胞的增殖。
