摘要 目的:探讨细胞毒性T淋巴细胞相关抗原4免疫球蛋白(Cytotoxic T Lymphocyte Antigen 4-i mmunoglobulin,CTLA4-Ig)与供体骨髓细胞联合应用诱导器官移植免疫耐受的可行性。方法:应用小鼠腹部心脏移植模型,实验组小鼠术中输注供体骨髓细胞,术后腹腔注射CTLA4-Ig。观察移植心脏存活时间,并进行病理分析,检测迟发性超敏反应(Delayed Type Hyper-sensitivity,DTH)。后期进行供体来源的皮肤移植并观察存活时间。结果:实验组移植心脏存活时间较对照组显著延长,达(79.3±23.7)d;排斥反应减轻,并产生了供体抗原特异性的免疫低反应性;后期进行的皮肤移植实验中,供体来源的皮肤存活时间也显著延长,达(23.0±3.1)d。结论:CTLA4-Ig与骨髓细胞联合应用可以显著延长移植物存活时间,诱导实体器官移植免疫耐受。两者具有明显的协同作用。 Objective: To explore the feasibility about the immune tolerance to murine cardiac allograft induced by combined application of Cytotoxic T Lymphocyte Antigen 4-immunoglobulin and Donor Bone Marrow cells.Methods: The recipients in the experimental group were infused with Donor-specific Bone Marrow Cells through the portal vein in the operation,and were infused with CTLA4-Ig into abdominal cavity postoperation.Then the survival time of the donor hearts was observed,histopathology of the donor heart allograft...
