丹参注射液诱导大鼠骨髓间充质干细胞分化为成软骨细胞的研究 被引量:23

摘要 目的:探讨复方丹参注射液体外诱导大鼠骨髓间充质干细胞(MSC)分化为成软骨细胞的作用。方法:大鼠MSC分离、培养、扩增、融合时传代,选用第3~5代细胞,分为3组,西药组加入高糖DMEM培养液(含地塞米松10-7mol/L、10%胎牛血清、TGF-β1 10μg/L、VitC 50mg/L),中药组加入高糖DMEM培养液(含0.5%的复方丹参注射液、10%胎牛血清),另设一空白组;倒置显微镜下逐日观察各组原代及传代细胞的生长情况及形态特征,诱导7d后,免疫组化法测定各组Ⅱ型胶原阳性率。结果:药物诱导处理后,各组均有部分细胞开始变为宽阔、平坦的多边形及多伪足伸展形细胞:Ⅱ型胶原的阳性率西药组和中药组比较无明显差异,二者均明显高于空白组(P<0.01)。结论:复方丹参注射液可体外诱导大鼠MSC分化为成软骨细胞。
