摘要
目的:用紫外分光光度法建立稀针嗜蓝孢孔菌多糖的定量分析方法。方法:采用苯酚-硫酸法,以葡萄糖含量为指标,用L9(34)正交试验法选择最佳显色条件,检测波长为488 nm。结果:苯酚、浓硫酸二因素对多糖含量有显著影响,以葡萄糖表示,本法在(3.223~12.89)μg.mL-1范围内,吸收度与浓度有良好的线性关系,平均回收率96.60%,RSD为1.71%。结论:本法快速、简便,灵敏度、重复性好,不受蛋白质干扰,亦可用于混有糖苷的粗多糖的测定。
Objective:To establish UV method to determine polysaccharide in Fomitiporia robusta(P.Karst.)
