猪圆环病毒2型ORF2和ORF1-2串联基因重组腺病毒对小鼠的免疫效果 被引量:1

摘要 为了研究含有猪圆环病毒2型(porcine circovirus type2,PCV2)基因的重组腺病毒作为基因工程疫苗的潜在应用价值,利用本实验室构建的PCV2的ORF2和ORF1-2串联基因的重组腺病毒对小鼠进行了免疫并对其免疫效果进行了研究。分别用表达ORF2和OFR1-2基因的重组腺病毒Ad-ORF2、Ad-ORF1-2经肌肉途径免疫Balb/c小鼠,对免疫后小鼠的血清抗体和脾淋巴细胞中各细胞亚群的比例及其增殖反应、自然杀伤活性和特异性杀伤活性进行了检测和比较。结果表明,用表达ORF2基因和ORF1-2串联基因的重组腺病毒Ad-ORF2、Ad-ORF1-2经肌肉途径免疫Balb/c小鼠后,随着免疫次数的增加,小鼠产生的针对PCV2ORF2基因的特异性抗体水平不断升高,第3次免疫后2周达到最高,但第3次免疫后4周有所下降;免疫重组腺病毒的试验组小鼠的脾淋巴细胞增殖活性、CTL杀伤活性和NK细胞杀伤活性与免疫空载体的对照组和免疫PBS的对照组比较均有显著的差异(P<0.05)。本研究为PCV-2基因工程疫苗的深入研究及应用奠定了基础。 In order to clarify the feasibility of adenovirus vector based porcine circovirus type 2,we investigated the immune responses induced by the adenoviruses carrying ORF2 and ORF1-2 gene on the bases of our previous reports.Balb/c mice were inoculated intramuscularly with recombinant adenovirus ORF2 and ORF1-2.The serum antibodies as well as the cellular characters were determinated.The results demonstrated that the serum antibody was arising with increase of the immunity times,the highest serum antibody appea...
