去卵巢对大鼠AGEs和LPO的影响及氨基胍和补肾方药的干预作用 被引量:1

摘要 目的研究去卵巢对大鼠晚期糖化终末产物(AGEs)和脂质过氧化物(LPO)的影响,探讨氨基胍和补肾方药的作用。方法40只9月龄雌性SD大鼠随机等分为卵巢假切除组(Sham组)、去卵巢组(OVX组)、氨基胍组(AG组)和补肾方药组(TSR组)。OVX组、AG组和TSR组大鼠切除双侧卵巢,Sham组大鼠切除腹腔等体积脂肪。药物干预16周后,用Y-电迷宫测试每组大鼠学习记忆能力,用ELISA法检测定大鼠大脑皮质、海马中AGEs含量,用ELISA法和荧光分光光度计2种方法同时检测血清和尿中AGEs含量,采用生化法检测LPO含量。结果与Sham组比较,OVX组逃避正确率明显降低(P<0.01),AG和TSR组逃避潜伏期缩短(P<0.05);与OVX组比较,AG和TSR组逃避正确率提高,逃避潜伏期缩短(P<0.05,P<0.01)。与Sham组比较,OVX组大鼠大脑皮质、海马组织AGEs含量没有明显升高(P均>0.05),血清AGEs(荧光性-AGEs和抗CML-AGEs)含量明显增加(P<0.05),血清、海马、大脑皮质中LPO含量明显增加(P<0.05);与OVX组和Sham组比较,AG组和TSR组大脑皮质AGEs含量均显著降低(P<0.05),血清中抗CML-AGEs含量明显增加(P<0.01),尿液中荧光性-AGEs和抗CML-AGEs含量明显增加(P<0.01),血清、海马、大脑皮质LPO含量均明显下降(P<0.05),AG组血清荧光性AGEs含量明显减少(P<0.05)。结论长期雌激素缺乏可造成大脑皮质、海马和血清LPO含量升高,血清AGEs含量升高,损害学习记忆能力;氨基胍和补肾方药都可通过减少体内AGEs和LPO沉积,恢复去卵巢大鼠的学习记忆能力。 Objective To explore the effects of ovariectomy on rats AGEs and LPO,and to discuss the effects of both aminoguanidine(AG)and TSR.Methods Forty Sprague-Dawley female rats(nine months)were randomly divided into four groups:Sham group,OVX group,AG group and TSR group.The ovaries were resected in OVX group,AG group and TSR group,and the same volume fats were resected in sham group.Learning and memory behavior of rats were tested in Y-electric maze after 16 weeks,and the content of AGEs and LPO in cerebral cor...
