摘要
以已制备的抗相思子毒素-a(abrin-a)的单克隆抗体(McAb)4G1和兔源多抗建立了abrin-a双抗体夹心ELISA法。abrin-a质量浓度在15.62~250.00μg/L的范围内,质量浓度的对数值与其D450值呈直线相关,回归方程为y=1.0467x+0.5277(R2=0.9992,n=5),检测灵敏度为7.81ng/mL。该法与相思子凝集素、蓖麻毒素(ri-cin)和蓖麻凝集素无交叉反应。经初步用于人工污染abrin-a香肠样品和自来水样品检测,回收率分别为92.0%和85.0%,变异系数(CV)<6%。表明该检测方法具有快速、特异、灵敏等特点,可望用于食品、饲料和饮用水中abrin-a检验。
Based on the prepared 4G1 monoclonal antibody(McAb) and rabbit serum anti-abrin-a,a double antibody sandwich enzyme-linked immunosorbent assay(ELISA) method for abrin-a was developed.There was line correlation between the logarithm values of C and D450 values,when the concentrations(C) of abrin-a varied from 15.62 to 250 mg/L.The detection equation was y=1.046 7x+0.527 7(R2=0.999 2,n=5),with a detection limit of 7.81 μg/L.No cross-reaction was found in abrus agglutinin,ricin and ricus agglutinin.It has been...
